ABSTRACT

The concentrations of interleukin 1 (IL-1) and interleukin 2 (IL-2) produced in the supernatants of peripheral blood mononuclear cell cultures from patients with advanced cancer were measured to identify some of the causes of the immunological impairment characteristic of malignant disease. Mononuclear cells obtained from 19 cancer patients were stimulated to produce IL-1 and IL-2 and compared with those of healthy controls. A severe reduction of both IL-1 and IL-2 activity was observed. There was no correlation between the lower number of OKT4+ cells observed in these patients and the levels of IL-2 production. The removal of monocytes did not bring IL-2 levels to normal. Impaired IL-2 production could not be restored to normal by addition of IL-1. These results suggest that exogenous IL-01 and IL-2 may be useful in cancer immunotherapy

ABSTRACT

In view of the rapid degradation of PAF in biological fluids, this study was designed to determine if late eosinophil infiltration induced in rats by PAF was deriived from its direct chemotatic action. A significant and selective 5-fold increase in the pleural eosinophil counts was detected 24 h after intrapleural PAF injection. The transfer of 6-h PAF washings to the pleural cavity of recipient rats also induced a 3-fold selective accumulation of eosinophils. The protein synthesis inhibitor cycloheximide abolished the pleural eosinophil migration and the generation of transferable chemotactic activity when administered to donor but not to recipient rats. These findings suggest that a secondary protein mediator accounts for the late eosinophil mobilization induced by PAF

ABSTRACT

O presente estudo foi realizado com a finalidade de analisar receptores de glicocorticóide em leucócitos mononucleares (MNL) de sangue periférico de indivíduos normais, näo tratados e de indivíduos normal aos quais foram administrados exogenamente os corticóides: prednisona e deflazacort. MNL de mulheres e homens normais apresentaram uma concentraçäo de receptores de glicocorticóide de 7,52 ñ 2,5fMol/10**6 cel e uma constante de dissociaçäo (Kd) de 9,5 ñ 2,3nM. Näo houve diferença em relaçäo ao sexo e as fases do ciclo menstrual. Em indivíduos tratados com predinisona, näo houve mudança nos parâmetros de ligaçäo. A administraçäo deflazacort determinou uma sensível reduçäo dos sítios de ligaçäo. Concluímos que certos glicocorticóides podem exercer um efeito regulatório sobre seus receptores

ABSTRACT

A resposta imunológica inespecífica foi avaliada "in vitro" em um grupo de crianças brasileiras de seis meses a cinco anos com má nutriçäo calórico-protéica moderada, näo-portadora de infecçäo, comparado com crianças eutróficas da mesma idade. Os desnutridos apresentaram depressäo da funçäo fagocitária, da resposta quimiotática e níveis séricos normais de complemento, indicando um problema intrínseco do leucocito mononuclear, o que pode ser uma das causas da frqüente associaçäo entre desnutriçäo e infecçäo
